چکیده

Development of new and improvement existing models long-term cultivation hematopoietic stem cells (HSC) cord blood progenitor is one the most important directions in modern cell biology, related to obtaining a sufficient amount tissue for experimental clinical use. The aim work was investigate functional activity from during on soft polyacrylamide gel substrate presence cytokine complex determine advantages this method maintaining hematopoiesis further use transplantation. To realize purpose, following methods were used: vitro hydrogel substrate, colony formation semi-liquid agar, cytological research methods, light inverted microscopy, statistical methods. Due together with complex, high proliferative observed, which reflected their expansion (5 weeks), as well colony-forming (521.5 ± 7.5.105 per explanted cells). Thus, stiffness must be taken into account immediate descendants. made along cytokines ensures due long- term support hematopoiesis.

Hemopoietic and Lymphoid Progenitor Cells in Human Umbilical Cord Blood

Human umbilical cord blood, which in the past was discarded with the placental tissue, provides a convenient source of fetal hemopoietic cells for scientific analysis and clinical use. Cord blood cells are immature compared to analogous populations in adult peripheral blood. Cord blood B lymphocytes display unique phenotypic and functional characteristics. The antigens CD1C, CD38, CD5, and CD23...

Isolation of Endothelial Progenitor Cells from Cord Blood and Induction of Differentiation by Ex Vivo Expansion

Endothelial progenitor cells (EPCs) have been reported to possess the capacity to colonize vascular grafts and hold promise for therapeutic neovascularization. However, limited quantities of EPCs have been the major factor impeding effective research on vasculoangiogenesis. In this study, cytokine and culture conditions necessary for the provision of large quantities of endothelial cells (ECs) ...

Ex Vivo Expansion of Hematopoietic Stem and Progenitor Cells from Umbilical Cord Blood

Transplantation of umbilical cord blood cells is currently widely used in modern cell therapy. However, the limited number of hematopoietic stem and progenitor cells (HSPCs) and prolonged time of recovery after the transplantation are significant limitations in the use of cord blood. Ex vivo expansion with various cytokine combinations is one of the most common approaches for increasing the num...

Effect of anti-CD52 antibody alemtuzumab on ex-vivo culture of umbilical cord blood stem cells

BACKGROUND Excessive maturation of hematopoietic cells leads to a reduction of long-term proliferative capability during cord blood (CB) expansion. In this study, we report the effects of anit-CD52 (Alemtuzumab, Campath) on both short- and long-term ex vivo expansion of CB hematopoietic stem cells (HSC) by evaluating the potential role of Alemtuzumab in preserving the repopulating capability in...
